Weight-loss peptides — the scientific background

One of the most talked-about areas in metabolism research is the family of peptides related to appetite and weight regulation — headed by agonists of the receptor GLP-1. GLP-1 is a hormone of the digestive system involved in the feeling of satiety, in slowing gastric emptying, and in regulating blood-sugar levels. To understand why this family is so interesting, it's worth getting to know the “gut-brain axis”: hormones like GLP-1 and GIP are released in the gut in response to eating and transmit to the brain signals of satiety and energy regulation. Compounds that mimic them prolong and strengthen these signals — hence the studied effect on appetite.

What's the difference between Ozempic, Mounjaro, and Retatrutide

The main difference is how many receptors each compound acts on. Semaglutide (known asOzempic andWegovy) is a single agonist — GLP-1 only. Tirzepatide (known asMounjaro) is a dual agonist — GLP-1 together with GIP. And the new generation, Retatrutide, is a triple agonist — GLP-1, GIP, and glucagon — acting on three pathways simultaneously. What's especially intriguing about Retatrutide is the glucagon pathway, which is linked to energy expenditure and not only to appetite suppression.

What the research shows

Results published in 2023 in the New England Journal of Medicine showed, in a Phase 2 trial, an average reduction of about 24% in body weight at the high dose, over 48 weeks — a result achieved in a controlled clinical-trial setting and under medical supervision, with an experimental, unapproved compound. As with other compounds in the family, side effects were also examined, mainly in the gastrointestinal system.

It's worth distinguishing between compounds already approved for supervised medical use in various countries (such as semaglutide and tirzepatide) and compounds that are still experimental (such as Retatrutide). An approved product has undergone control and approval processes, whereas an experimental compound is still in the research stages — and some of the information about it, including its long-term safety profile, is still being studied. In any case, the products here are sold as research materials only.
